How Fluconazole 150 mg Tablets Work

Fluconazole is an artificial triazole antifungal drug which acts by inhibiting the fungal cytochrome P450-dependent 14α-demethylase enzyme (CYP51A1). This enzyme plays a key role in the production of ergosterol from lanosterol. Ergosterol is the major sterol in the fungal cell wall. Fluconazole inhibits the function of the fungal cytochrome, resulting in a reduction of ergosterol content in the fungal wall. This reduces the integrity of the fungal cell wall, increases membrane permeability, causes loss of cellular contents, and hinders the growth of fungi. Fluconazole has higher specificity for the fungal CYP51 enzyme than the mammalian CYP51 enzyme.

Dosage and Administration

Fluconazole 150 mg Tablets should only be administered orally. In the case of uncomplicated vulvovaginal candidiasis, the recommended dose of one 150 mg tablet orally is normally used. The dose for other indications (oropharyngeal candidiasis, systemic infections) may differ — it is recommended to adhere to the doctor’s recommendations exactly. It can be taken irrespective of eating. Self-treatment of recurrence or complicated infections is not recommended.

Side Effects

• Fluconazole is an inhibitor of CYP2C9 and CYP3A4 — considerable drug interactions with warfarin, oral hypoglycemics, tacrolimus, phenytoin, and numerous other drugs
• Tell the prescribing physician about all other medications currently being used before taking
• Monitor liver function tests in cases where repeated or long-term therapy is required
• Contraindicated in pregnancy – Fluconazole is teratogenic; a single dose of 150 mg is hazardous in pregnancy
• Avoid use while breast-feeding unless directed by physician
• Store out of reach of children

Contraindications

Fluconazole 150 mg Tablets are contraindicated in known hypersensitivity to Fluconazole or other azole antifungals. Contraindicated with concurrent terfenadine, astemizole, cisapride, pimozide, or quinidine (risk of fatal cardiac arrhythmias). Avoid in pregnancy.
